I have received a conditional offer for E12 physiotherapy next year but did not study physics (assumed knowledge) for HSC.
Does the mean I won't get the place even if I got the E12 ATAR?

It's assumed, but not strictly required. This means they'll expect you to know some content from HSC physics, but you can still get into the course without actually having done HSC physics.
So don't worry about it. I think the uni runs HSC physics bridging courses during the post-HSC break. Honestly, you could probably just self-study preliminary/HSC physics during the post-HSC break and you should be fine.
