Do you have to have a question for your critical response?

Not particularly. Although, I, myself am not doing a critical response, others seem to base theirs on an idea, or concept. However, it does seem easier to respond to and fulfil the criteria thoroughly using a question: Asian influences in literature vs. How has Asian culture found its systemic roots in contemporary literature? Being more specific in your question will allow your response to have a stronger and more multi-dimensional underlying point.

That all being said, this is just my opinion; do what you think will be good.

No. You obviously need a purpose behind your response, but not a formalised question per se.
