Take a leap of faith and tell her! You've got the world to gain... and well, hopefully experience minimal losses.
I mean... if you're afraid of risking your friendship, then just think about if she gets a boyfriend. Pretty sure you'll get hurt (as you are harbouring feelings for her) and in the end your friendship will still get tinted.
Love is like a stock market (dear Lord, I hope my limited economics knowledge will help me pull of this metaphor). Sometimes, you just gotta risk your money and invest in a profitable looking business/ stock. Likewise, you need to risk your friendship so that you can potentially get something more from it (a relationship if that is what you are after). In both the business world and love: it's all about the guts

Biggest problem is that it's his best friend which is no small deal, honestly man just give her hints, (make sure you tell the difference between shy and straight out rejecting them) , you'll know if she takes on any of them or not, biggest thing you got going for you is that since she's your best friend getting comfortable is easy, and you can always do a quick "sorry" after trying to be "more" comfortable with her
